"Rose with prickles” is a symbol of adversity and sacrifice but the molecular bases patterning prickle remains poorly explored. Here we generated a high-quality reference genome assembly for prickleless Rosa wichuraiana ‘Basye’s Thornless’ (BT), and identified genetic elements relating to stem prickle patterning. BT genome harbors a significant high proportion of Copia-type LTR (62.74% of LTRs) and diverges remarkably from that of R. chinensis Old Blush’, a founder genotype for rose domestication. Three negatively selected and fixed QTL were identified underlying BT prickleless. Differentially expressed genes in QTL were enriched for water-use-related genes, suggesting that prickleless might hitchhike the adaptation to moisture environments. While the prickle-related gene-regulatory-network (GRN) was highly conserved, expression variation of key candidate genes associated well with prickle-patterning divergence in roses. Our efforts provide fundamental resources and insights for Rosaceae genome evolution and molecular basis of key traits, thus should benefit rose improvement.

Seed longevity (i.e., the inherent lifespan of a seed) is a polygenic feature that demands the coordination of multiple biological processes, and that shapes the persistence of seeds in the soil and the time they can be stored ex situ. Recent studies on this trait highlight the role of the antioxidant and biomolecule repair systems and the formation of the seed coat. However, the genetic basis behind their contribution has not yet been fully described or corroborated. Here, Renard et al. perform a Genome-Wide Association Study (GWAS) to identify the genes responsible for the differences in the seed longevity of 270 ecotypes of Arabidopsis thaliana. Seeds were exposed to high humidity, high temperature or elevated oxygen pressure conditions (or a combination of these) in order to accelerate the loss of their viability. The results of these assays were correlated with the genomic data available on public databases. After this, 20 candidate genes were validated through a mutant analysis, and 12 had a significant effect in longevity. Most genes were related to enzymes that participate in the metabolism of reactive oxygen species, such as NADPH oxidases (RBOHs) and dehydroascorbate reductases (DHAR1). A cytochrome hydroxylase (CYP86A8) and several transcription factors related to seed coat development also promoted longevity. This paper provides a promising starting point for future research about the mechanisms behind seed longevity.  (Summary by Carlos A. The CLAND and Saclay Plant Sciences (SPS) networks are launching a call which aims at fostering interactions and common research projects between the two communities. Through this call, CLAND and SPS will cofund one PhD project involving teams from both networks.

To be noted: The CLAND-SPS Meeting, initially planned on April 21st and postponed because of the Covid19 situation, will be organized in 2021 and a second call for a common CLAND-SPS PhD project will be launched following the meeting.

CLAND (https://cland.lsce.ipsl.fr) is a Convergence Institute funded by the French government in 2017 to perform in the next decade the research urgently needed on land-management solutions for managing the ecological and energy transitions of the 21st century. It aims to support the activity of 200 researchers in 12 teams and develop a graduate program across Master and Doctoral schools of the University of Paris-Saclay, in partnership with European and international institutes.

The Saclay Plant Sciences network (SPS, www.saclayplantsciences.fr) was created through the French “Investments for the Future” program in 2011. It gathers around 50 research teams working on plant sciences in five laboratories from the southern Paris area, as well as several teaching programs, among which a Master program and a Doctoral School entirely dedicated to plant sciences.

Conditions of the call
The projects must involve at least one CLAND team and one SPS team.

The funding granted to the successful proposal will cover the salary of the PhD candidate over the three years of PhD and up to 6 000 euros of running costs (the use of this envelope must de detailed in the project).

Projects have to be written in ENGLISH.

The Project Leaders of the selected project commits:
- to providing the curriculum vitae of the future PhD student as soon as possible,
- to providing any additional information requested by CLAND or SPS about the project, during or after the project (notably as part of the CLAND and SPS activity reports).

Editor's view: The parasitic plant Striga hermonthica germinates when it senses strigolactones (SLs). By expressing Striga SL receptors in Arabidopsis, the authors show that the SL pathway can regulate germination independently of gibberellin signalling.


Abstract: "Parasitic plant infestations dramatically reduce the yield of many major food crops of sub-Saharan Africa and pose a serious threat to food security on that continent1. The first committed step of a successful infestation is the germination of parasite seeds primarily in response to a group of related small-molecule hormones called strigolactones (SLs), which are emitted by host roots2. Despite the important role of SLs, it is not clear how host-derived SLs germinate parasitic plants. In contrast, gibberellins (GA) acts as the dominant hormone for stimulation of germination in non-parasitic plant species by inhibiting a set of DELLA repressors3. Here, we show that expression of SL receptors from the parasitic plant Striga hermonthica in the presence of SLs circumvents the GA requirement for germination of Arabidopsis thaliana seed. Striga receptors co-opt and enhance signalling through the HYPOSENSITIVE TO LIGHT/KARRIKIN INSENSITIVE 2 (AtHTL/KAI2) pathway, which normally plays a rudimentary role in Arabidopsis seed germination4,5. AtHTL/KAI2 negatively controls the SUPPRESSOR OF MAX2 1 (SMAX1) protein5, and loss of SMAX1 function allows germination in the presence of DELLA repressors. Our data suggest that ligand-dependent inactivation of SMAX1 in Striga and Arabidopsis can bypass GA-dependent germination in these species."

The 1,000 Plants (1KP) initiative was the first large-scale effort to collect next-generation sequencing (NGS) data across a phylogenetically representative sampling of species for a major clade of life, in this case the, or green plants. As an international multidisciplinary consortium, we focused on plant evolution and its practical implications. Among the major outcomes were the inference of a reference species tree for green plants by phylotranscriptomic analysis of low-copy genes, a survey of paleopolyploidy (whole-genome duplications) across the , the inferred evolutionary histories for many gene families and biological processes, the discovery of novel light-sensitive proteins for optogenetic studies in mammalian neuroscience, and elucidation of the genetic network for a complex trait (C photosynthesis). Altogether, 1KP demonstrated how value can be extracted from a phylodiverse sequencing data set, providing a template for future projects that aim to generate even more data, including complete de novo genomes, across the tree of life.

A major change to U.S. regulation of biotech will exempt some gene-edited plants from government oversight. The new policy, published in the Federal Register today, also calls for automatic approval of variations of established kinds of genetically modified (GM) crops, easing their path to market.

Industry groups are welcoming the new rule, whereas opponents are decrying the reduction of government oversight.

“The main good thing is that it will allow certain aspects of gene editing to move forward,” says Kent Bradford, a plant geneticist at the University of California, Davis. If researchers use gene editing to design a plant that could have been bred conventionally, the new plant will be exempt from regulation. But anything else—such as moving a gene between species or rewiring metabolism—will still require a regulatory review.

 

 

The gist of the shift is that the U.S. Department of Agriculture’s (USDA’s) Animal and Plant Health Inspection Service (APHIS) will now focus on new traits themselves rather than the technology used to create them, a change of approach that plant scientists have long wanted. Several reviews by the National Academy of Sciences have concluded that the risk that GM plants will become weeds is generally low, and that molecular tools typically don’t pose new risks compared with traditional plant breeding techniques.

 

The change to the USDA regulations began during the Obama administration. The Trump administration released draft rules in January 2017, then withdrew them 9 months later. (In its new rule, USDA says it received comments that the previous version would be “too burdensome and had the potential to stifle innovation.”) Last summer, USDA released a revised rule for public comment, which it has now finalized.

An engineered plant won’t be regulated if it contains minor changes—a change to a pair of amino acid bases or a deletion of a chunk of DNA—that would create a trait that could have been made through traditional breeding. For example, molecular biologists can snip disease resistance genes from various parts of a plant’s genome and gather them into one stretch of DNA, allowing breeders to easily incorporate all the genes into one variety. The end product will be the same as what might be created by breeders, but gene editing can save years of effort, Bradford says. “This makes everything hugely easier.” But he’s frustrated that the exemption doesn’t cover more substantive changes or moving genes between closely related plants, such as peppers and tomatoes, which can’t be crossed with conventional breeding.

 

Another change will make it easier to create minor variations of GM crops, such as tailoring them for different climates. Before, companies had to ask APHIS to evaluate the risk of any new GM crop they wished to commercialize, even if it had been altered in the same way as crops already approved. Now, APHIS won’t regulate new varieties of an already approved GM crop.

If a company isn’t sure whether its new crop is exempt, it can consult APHIS. Regulators will look at the proposed trait in comparable plants to see whether there’s any risk. The agency expects that such reviews could be done in 2 to 3 months for familiar plants. APHIS estimates that about 1% of plants might not qualify for an exemption, or deregulation after an initial review. That’s welcome news to plant developers. “The new process established by this rule is expected to lead to lower regulatory costs and timeframes for the development of new plant varieties,” the American Soybean Association said in a statement.

APHIS says it wants to keep up with evolving science and technology. Research institutes, companies, or other stakeholders can ask APHIS to expand the exemptions, a process that would involve public comment. “A clear and transparent, science-based process for these future exemptions will be important to support continuing innovation,” said the American Seed Trade Association in a statement.

 

Some groups are concerned that companies won’t have to notify USDA of biotech crops they will bring to market that are exempt from regulation. “The result is that government regulators and the public will have no idea what products will enter the market and whether those products appropriately qualified for an exemption from oversight,” Gregory Jaffe of the Center for Science in the Public Interest said in a statement.

The Biotechnology Innovation Organization (BIO), a major trade group, appeared to encourage companies make public such releases. “BIO encourages increased openness about products entering the marketplace,” it said in a statement.

Most of the new changes will go into effect on 5 April 2021.

Abstract
Trace metal elements are essential for plant growth but become toxic at high concentrations, while some non-essential elements, such as Cd and As, show toxicity even in traces. Thus, metal homeostasis is tightly regulated in plants. Plant species colonising metalliferous soils have evolved mechanisms to hypertolerate metals and, in rare cases, can hyperaccumulate them in excess amounts in their shoots. The molecular mechanisms of metal hypertolerance and hyperaccumulation are likely derived from alterations in the basic mechanisms involved in general metal homeostasis. Genes involved in metal transport, synthesis of metal chelators and oxidative stress responses are constitutively and highly expressed in metal hypertolerant and hyperaccumulator species. Plant specialized metabolites and cell wall components have been proposed as major players in these mechanisms. In addition, the high intra-specific natural variation of metal hypertolerance and hyperaccumulation suggests that various molecular mechanisms might be involved in the evolution of these traits. To date, the potential of wild plant populations as systems to study metal tolerance and hyperaccumulation has not been fully exploited. The advent of next-generation sequencing (NGS) has enabled the study of non-model species, providing an opportunity to study natural populations and new tolerant and/or hyperaccumulating species, and will provide new insights into metal tolerance and hyperaccumulation. In this review we highlight background knowledge about metal tolerance and hyperaccumulation in plants and the current state-of-the-art techniques to study and identify the underlying mechanisms of metal hypertolerance and hyperaccumulation. We also outline for the reader the importance of the multidisciplinarity of this research field and how the integration of multiomic approaches will benefit facing the future scientific challenges.

Plants produce a huge diversity of specialized metabolites (SM) throughout their life cycle that play important physiological and ecological functions. SM can protect plants and seeds against diseases, predators, and abiotic stresses, or support their interactions with beneficial or symbiotic organisms. They also have strong impacts on human nutrition and health. Despite this importance, the biosynthesis and biological functions of most of the SM remain elusive and their diversity and/or quantity have been reduced in most crops during domestication. Seeds present a large number of SM that are important for their physiological, agronomic, nutritional or industrial qualities and hence, provide interesting models for both studying biosynthesis and producing large amounts of specialized metabolites. For instance, phenolics are abundant and widely distributed in seeds. More specifically, flavonoid pathway has been instrumental for understanding environmental or developmental regulations of specialized metabolic pathways, at the molecular and cellular levels. Here, we summarize current knowledge on seed phenolics as model, and discuss how recent progresses in omics approaches could help to further characterize their diversity, regulations, and the underlying molecular mechanisms involved.

Abstract: "Seed development, which involves mainly the embryo, endosperm and integuments, is regulated by different signaling pathways, leading to various changes in seed size or seed weight. Therefore, uncovering the genetic and molecular mechanisms of seed development has great potential for improving crop yields. The phytohormone auxin is a key regulator required for modulating different cellular processes involved in seed development. Here, we provide a comprehensive review of the role of auxin biosynthesis, transport, signaling, conjugation, and catabolism during seed development. More importantly, we not only summarize the research progress on the genetic and molecular regulation of seed development mediated by auxin but also discuss the potential of manipulating auxin metabolism and its signaling pathway for improving crop seed weight."

The process by which plant cells expand and gain shape has presented a challenge for researchers. Current models propose that these processes are driven by turgor pressure acting on the cell wall. Using nanoimaging, we show that the cell wall contains pectin nanofilaments that possess an intrinsic expansion capacity. Additionally, we use growth models containing such structures to show that a complex plant cell shape can derive from chemically induced local and polarized expansion of the pectin nanofilaments without turgor-driven growth. Thus, the plant cell wall, outside of the cell itself, is an active participant in shaping plant cells. Extracellular matrix function may similarly guide cell shape in other kingdoms, including Animalia.

Plus de 700 directrices et directeurs de laboratoires de recherche contestent, dans une tribune au « Monde », les critères qui président à l’élaboration de la loi de programmation pluriannuelle de la recherche. Ils préconisent de « renforcer les collectifs » plutôt que de promouvoir « une infime élite œuvrant au profit d’une infime partie des savoirs »

 

Tribune. Faute de tenir les engagements budgétaires pris par l’Etat à l’occasion de l’agenda de Lisbonne (2000), le service public de recherche et d’enseignement supérieur décroche. Son indépendance et sa stabilité sont ébranlées par des pratiques d’évaluation, de management et de gestion empruntées au secteur privé. Les politiques publiques favorisent ainsi les contrats courts et précaires au détriment des emplois pérennes, elles promeuvent la compétition contre la coopération et enjoignent de trouver des résultats rentables aux dépens de la liberté de chercher et de son incertitude radicale.

 

Le 1er février, confirmant le projet d’élaboration d’une loi de programmation pluriannuelle de la recherche, le premier ministre déclarait : « Gouverner, c’est choisir, et choisir, c’est renoncer ; nous devons donc faire des choix et décider ce qui est le plus important. »On comprend qu’il ne faudra garder que les « meilleurs » d’entre nous. Mais selon quels critères ? Ceux qui obtiennent des prix internationaux ou des labellisations d’« excellence », déposent des brevets, obtiennent de très gros financements européens et font des recherches à haut « facteur d’impact » ?

 

Soumettant la recherche à une pensée utilitariste et comptable, le management public mesure aujourd’hui la performance à court terme et ne considère le fruit du travail scientifique que par les seuls travaux à haute visibilité et à rentabilité immédiate. Pourtant chacun sait que la recherche ne peut être que le fait d’une vaste communauté, « stars » comprises : aucun « génie » n’existe sans un collectif stable, compétent et à l’abri de la précarité.

Des postes précaires de compensation de plus en plus nombreux

L’application généralisée et systématique des critères de la « performance », dont on connaît pourtant bien les défauts et les travers, constitue un danger majeur qui met notamment en péril notre indépendance vis-à-vis des financements privés et internationaux. Que se passera-t-il quand les chercheuses et les chercheurs travaillant dans les domaines de l’énergie, de l’économie,de l’agriculture ou de la pharmacie ne seront plus financés que par le secteur privé ou des intérêts sectoriels ? Les sciences où les brevets sont rares, qui ne nécessitent pas de gros équipements et dont les travaux profitent davantage à l’intelligence collective et au progrès social sont-elles amenées à disparaître ?

  • As abscisic acid (ABA) receptors, PYR1/PYL/RCAR (PYLs) play important roles in ABA‐mediated seed germination, but the regulation of PYLs in this process, especially at the transcriptional level, remains unclear.
  • In this study, we found that expression of 11 of 14 PYL s changes significantly during seed germination and is affected by exogenous ABA. Two PYLs, PYL11 and PYL12, both of which are expressed specifically in mature seeds, positively modulate ABA‐mediated seed germination.
  • However, ABI5 was found to modulate the PYL11‐ and PYL12‐mediated ABA response. In the abi5‐7 mutant, ABA hypersensitivity caused by PYL11 and PYL12 overexpression was totally or partially blocked. By contrast, ABI5 regulates the expression of PYL11 and PYL12 by directly binding to their promoters. Moreover, the expression of eight other PYL s is also affected during the germination of abi5 mutants. Promoter analysis revealed that an ABI5‐binding region is present next to the TATA box or initiator box.
  • Together, our data demonstrate the role of PYL11 and PYL12 in seed germination. In addition, the identification of PYL s as targets of ABI5 reveals a role of ABI5 in the feedback regulation of ABA‐mediated seed germination.

Genomic imprinting is an epigenetic phenomenon altering the activity of parental alleles depending on their parent-of-origin. In flowering plants, imprinting is mainly confined to the endosperm, an embryo supportive tissue similar to the placenta in mammals. Epigenetic imprints are established during gamete formation; however, the determining factors for imprinting establishment remain obscure. We identified the MADS-box transcription factor PHERES1 as master regulator of imprinted gene expression in the flowering plant Arabidopsis thaliana, especially of paternally expressed genes, which have been previously implicated in endosperm development. Control of imprinted gene expression by PHERES1 is mediated by parental asymmetry of epigenetic modifications in PHERES1 DNA-binding sites, conferring different accessibilities to maternal and paternal alleles. Importantly, the DNA-binding motifs used by PHERES1 to access gene promoters are carried by RC/Helitron transposable elements, providing an example of molecular domestication of these elements. Thus, transposable elements are intrinsically linked to imprinting and endosperm development, not only by enforcing specific epigenetic landscapes, but also by serving as important sources of cis-regulatory elements.

Abstract: "Arabidopsis thaliana ABSCISIC ACID INSENSITIVE3 (ABI3) is a transcription factor in the B3 domain family. ABI3, along with B3 domain transcription factors LEAFY COTYLEDON2 (LEC2) and FUSCA3 (FUS3), and LEC1, a subunit of the CCAAT box binding complex, form the so‐called LAFL network to control various aspects of seed development and maturation. ABI3 also contributes to the abscisic acid (ABA) response. We report on chromatin immunoprecipitation‐tiling array experiments to globally map binding sites for ABI3. We also assessed transcriptomes in response to ABI3 by comparing developing abi3‐5 and wild type seeds and combine this information to ascertain direct and indirect responsive ABI3 target genes. ABI3 can directly induce and repress its target genes’ transcription and some intriguing differences exist in cis motifs between these groups of genes. Directly regulated targets reflect ABI3’s roles in seed maturation, desiccation tolerance, entry into a quiescent state and longevity. Interestingly, ABI3 directly represses a gene encoding a microRNA (MIR160B ) that targets AUXIN RESPONSE FACTOR (ARF )10 and ARF16 that are involved in establishment of dormancy. In addition, ABI3, like FUS3, regulates genes encoding MIR156 but while FUS3 only induces genes encoding this product, ABI3 induces these genes during early stages of seed development, but represses these genes during late development. The interplay between ABI3, the other LAFL genes, and the VP1/ABI3‐LIKE (VAL ) genes that are involved in the transition to seedling development are examined and reveal complex interactions controlling development."

Abstract: "Organ size and shape are precisely regulated to ensure proper function. The four sepals in each Arabidopsis thaliana flower must maintain the same size throughout their growth to continuously enclose and protect the developing bud. Here we show that DEVELOPMENT RELATED MYB-LIKE 1 (DRMY1) is required for both timing of organ initiation and proper growth, leading to robust sepal size in Arabidopsis. Within each drmy1 flower, the initiation of some sepals is variably delayed. Late-initiating sepals in drmy1 mutants remain smaller throughout development, resulting in variability in sepal size. DRMY1 focuses the spatiotemporal signalling patterns of the plant hormones auxin and cytokinin, which jointly control the timing of sepal initiation. Our findings demonstrate that timing of organ initiation, together with growth and maturation, contribute to robust organ size."

Genome editing by site‐directed nucleases (SDNs) such as TALEN or Cas9 is a versatile tool that generates variations in the recipient genome at specific target sites. SDNs produce a sequence‐specific DNA break that is repaired by the plant's natural DNA repair mechanisms; as the repair is inherently imperfect, it results in target‐site variants. Depending on the type of approach, we can distinguish between three types of alterations. SDN‐1 introduces base‐pair changes or small insertions/deletions without addition of foreign DNA. The exact change cannot be predetermined and is quasi random at the target site. SDN‐2 uses a small DNA template to generate a specific change by homologous recombination. SDN‐3 inserts larger DNA elements of foreign origin using a similar approach as SDN‐2 (ref. 1); the introduction of larger pieces of DNA is typically considered as transgenic. Many countries have now adapted their biosafety legislation based on this classification of SDN‐induced variants (Fig 1).

The plant immune system is broadly characterised as a cell autonomous response system; in general, all plant cells are capable of pathogen perception and response. However, plasma membrane-lined pores that connect neighbouring cells called plasmodesmata are regulated during immune responses, identifying that cell-to-cell connectivity and communication is a component of immune signalling. Plasmodesmata close in response to the perception of pathogens, mediated by specialised immune signalling cascades located at plasmodesmata. Antagonistically, pathogens can also regulate plasmodesmata to gain access to non-infected cells. Our research investigates how plasmodesmata and cell-to-cell connectivity are regulated within, and underpin, both immunity and infection.

Functional divergence after gene duplication plays a central role in plant evolution. Among cereals, only Hordeum vulgare (barley), Triticum aestivum(wheat) and Secale cereale (rye) accumulate delphinidin‐derived (blue) anthocyanins in the aleurone layer of grains, whereas Oryza sativa (rice), Zea mays (maize) and Sorghum bicolor (sorghum) do not. The underlying genetic basis for this natural occurence remains elusive. Here, we mapped the barley Blx1 locus involved in blue aleruone to an approximately 1.13 Mb genetic interval on chromosome 4HL, thus identifying a trigenic cluster named MbHF35 (containing HvMYB4H, HvMYC4H and HvF35H). Sequence and expression data supported the role of these genes in conferring blue‐coloured (blue aleurone) grains. Synteny analyses across monocot species showed that MbHF35 has only evolved within distinct Triticeae lineages, as a result of dispersed gene duplication. Phylogeny analyses revealed a shared evolution pattern for MbHF35 in Triticeae, suggesting that these genes have co‐evolved together. We also identified a Pooideae‐specific flavonoid 3′,5′‐hydroxylase (F3′5′H) lineage, termed here Mo_F35H2, which has a higher amino acid similarity with eudicot F3′5′Hs, demonstrating a scenario of convergent evolution. Indeed, selection tests identified 13 amino acid residues in Mo_F35H2 that underwent positive selection, possibly driven by protein thermostablility selection. Furthermore, through the interrogation of barley germplasm there is evidence that HvMYB4H and HvMYC4H have undergone human selection. Collectively, our study favours blue aleurone as a recently evolved trait resulting from environmental adaptation. Our findings provide an evolutionary explanation for the absence of blue anthocyanins in other cereals and highlight the importance of gene functional divergence for plant diversity and environmental adaptation.

Stakeholders working within the potato sector in Uganda are confident about the wide adoption of a new bioengineered late blight resistant potato. Dubbed the “3R Victoria” potato, the yet to be released variety could help over 300,000 smallholder farmers in Uganda achieve higher yields at a lower cost with less exposure to chemicals.

 

At an expert consultative meeting held in Uganda, stakeholders discussed critical issues that could affect the adoption of 3R Victoria. After long discussion, the group arrived at an estimated 40-50% adoption rate for the new variety 15 years after its release.

Dr. Charles Mugoya, the chairperson of the National Biosafety Committee, explained the variety’s potential benefits and constraints. For instance, using 3R Victoria, farmers will have to spend less money on fungicides. Traders will have to use clear labelling when purchasing and trading the 3R Victoria and they will also have to keep 3R Victoria tubers separated from other tubers. Seed companies selling 3R Victoria seeds will be registered and inspected for maintaining good stewardship, separate production lines, and packaging basic seeds in GMO-labelled bags.

Important factors that could influence the adoption of the 3R Victoria include availability of quality seed, its market value, and market presence among potential buyers. This is according to the workshop participants who represented key actors within the potato sector in Uganda.

Abstract: "The timing of the germination of seeds is highly responsive to inputs from the environment. Temperature plays a key role in the control of germination, with low temperatures acting to stimulate this developmental transition in many species. In Arabidopsis, extensive gene expression changes have been reported at the whole seed level in response to cold, while much less is known about their spatial distribution across the diverse cell types of the embryo. In this study we examined the spatiotemporal patterns of promoter activity and protein abundance for key gibberellic acid (GA) and abscisic acid (ABA) factors which regulate the decision to germinate both during a time course of germination and in response to cold. Low temperature stimulated the spatial relocalization of these factors to the vasculature. The response of these seeds to dormancy-breaking stratification treatments therefore stimulates the distribution of both positive (GA) and negatively acting (ABA) components to this same cell type. This altered spatial pattern persisted following the transfer of seeds to 22°C, as well as after their rehydration, indicating that this alteration is persistent. These observations suggest that the vasculature plays a role in the low temperature-mediated stimulation of germination in this species, while novel cell types are recruited to promote germination in response to stratification."

Abstract: "Seed germination is a complex biological process controlled by various regulators, including phytohormones. Among these, abscisic acid and gibberellic acid inhibit and promote seed germination, respectively. Many studies have addressed the biological roles of auxin in plant growth and development, but very few have considered its role in seed germination. Here, we identified a novel function of the auxin signaling repressor Aux/IAA8 during seed germination. The IAA8 loss-of-function mutant iaa8-1 exhibited delayed seed germination. The phenotype of iaa8-1 was restored by ectopic expression of IAA8. Interestingly, IAA8 accumulated to high levels during seed germination, which was achieved not only by increased protein synthesis but also by the stabilization of IAA8 protein. We also showed that IAA8 down-regulates the transcription of ABSCISIC ACID INSENSITIVE3 (ABI3), a negative regulator of seed germination. Our study, thus strongly suggest that the auxin signaling repressor IAA8 acts as a positive regulator of seed germination in Arabidopsis thaliana."
