maintain tolerance

Gilbert C FAURE's insight:

These data describe an immune cell pathway that originates in the CNS and is capable of dampening anti-CNS immune responses in the periphery. Furthermore, these data provide insight into how fingolimod treatment might exacerbate CNS neuroinflammation in some cases and suggest that focal therapeutic interventions, outside the CNS have the potential to selectively modify anti-CNS immunity.

Natural Killer Cells and Neuroblastoma: Tumor Recognition, Escape Mechanisms, and Possible Novel Immunotherapeutic Approaches | NK Cell Biology

Natural Killer Cells and Neuroblastoma: Tumor Recognition, Escape Mechanisms, and Possible Novel Immunotherapeutic Approaches | NK Cell Biology | NeuroImmunology | Scoop.it
Neuroblastoma (NB) is the most common extra-cranial solid tumor of childhood and arises from developing sympathetic nervous system. Most primary tumors localize in the abdomen, the adrenal gland or lumbar sympathetic ganglia.

Conclusion

Progress in the understanding of NB cell biology will allow a more accurate stratification of patients, thus reducing toxic side effects of aggressive therapy in low-risk patients. High-risk patients who currently have a dismal prognosis could benefit from multidisciplinary therapeutic protocols that include novel NK cell-based immunotherapeutic strategies. The latter will take advantage of our knowledge about the presence/absence of NB-associated ligands interacting with activating/inhibitory receptors expressed by NK cells. Moreover, it should also take into account the multiple immunomodulatory strategies set up by NB and various immune cell types to impair the recruitment and activation of NK cells in the tumor microenvironment. The encouraging results emerged from haploidentical hematopoietic cell transplantation in pediatric hematological malignancies, might strongly motivate a re-evaluation of transplant approaches in the therapy of high-risk NB patients.

The diagnosis of multiple sclerosis and the vari... [J Autoimmun. 2014] - PubMed - NCBI

PubMed comprises more than 23 million citations for biomedical literature from MEDLINE, life science journals, and online books. Citations may include links to full-text content from PubMed Central and publisher web sites.
Gilbert C FAURE's insight:

Several variants of MS (and CNS demyelinating syndromes in general) have been nowadays defined in an effort to increase the diagnostic accuracy, to identify the unique immunopathogenic profile and to tailor treatment in each individual patient. These include the initial events of demyelination defined as clinically or radiologically isolated syndromes (CIS and RIS respectively), acute disseminated encephalomyelitis (ADEM) and its variants (acute hemorrhagic leukoencephalitis-AHL, Marburg variant, and Balo's concentric sclerosis), Schilder's sclerosis, transverse myelitis, neuromyelitis optica (NMO and NMO spectrum of diseases), recurrent isolated optic neuritis and tumefactive demyelination.

New blood cells fight brain inflammation

Hyperactivity of our immune system can cause a state of chronic inflammation. If chronic, the inflammation will affect our body and result in disease. In the devastating disease multiple sclerosis, hyperactivity of immune cells called T-cells induce chronic inflammation and degeneration of the brain. Researchers at BRIC, the University of Copenhagen, have identified a new type of regulatory blood cells that can combat such hyperactive T-cells in blood from patients with multiple sclerosis.

Page Details (Find out How Imaris was used to quantify Morphology changes of Choroid Plexus after Immune Cells invade the CNS http://t.co/FueBPaAqiI)...
Gilbert C FAURE's insight:

The researchers are using the experimental autoimmune encephalomyelitis (EAE) animal model of multiple sclerosis to study the choroid plexus. They determined the gene expression patterns in the separate choroid plexus compartments in situ using laser capture microdissection and global RNA profiling. They turned to Imaris for the 3D morphological analysis because they had previously used it in a study that quantified capillary density and extent of branching within cerebral microvasculature.
