Frontiers | Enhanced Mucosal Antibody Production and Protection against Respiratory Infections Following an Orally Administered Bacterial Extract | Pulmonary Medicine

Secondary bacterial infections following influenza infection are a pressing problem facing respiratory medicine. Although antibiotic treatment has been highly successful over recent decades, fatalities due to secondary bacterial infections remain one of the leading causes of death associated with influenza. We have assessed whether administration of a bacterial extract alone is sufficient to potentate immune responses and protect against primary infection with Influenza, and secondary infections with either Streptococcus pneumoniae or Klebsiella pneumoniae in mice. We show that oral administration with the bacterial extract, OM-85, leads to a maturation of dendritic cells and B cells characterised by increases in MHC II, CD86, CD40 and a reduction in ICOSL. Improved immune responsiveness against Influenza virus reduced the threshold of susceptibility to secondary bacterial infections, and thus protected the mice. The protection was associated with enhanced polyclonal B cell activation and release of antibodies that were effective at neutralising the virus. Taken together, these data show that oral administration of bacterial extracts provides sufficient mucosal immune stimulation to protect mice against a respiratory tract viral infection and associated sequelae.

Gilbert C FAURE's insight:

Human holobiomes are networks of mutualistic interactions between human cells and complex communities of bacteria and fungi that colonize the human body. The immune system must tolerate colonization with commensal bacteria and fungi but defend against invasion by either organism. Molecular ecological surveys of the human prokaryotic microbiota performed to date have revealed a remarkable degree of bacterial diversity and functionality. However, there is a dearth of information regarding the eukaryotic composition of the microbiota. In this review, we describe the ecology and the human niches of our fungal “fellow travelers” in both health and disease, discriminating between passengers, colonizers, and pathogens based on the interaction of these fungi with the human immune system. We conclude by highlighting the need to reconsider the etiology of many fungal and immune-related diseases in the context of the crosstalk between the human system and its resident microbial communities.

non throat swabs

Yolken and colleagues stumbled upon the algae virus unexpectedly while analyzing the microbial population of the throats of healthy humans for a non-related study. Investigators obtained throat swabs and performed DNA analysis designed to detect the genetic footprints of viruses and bacteria. To their surprise, the researchers say, they discovered DNA matching that of Acanthocystis turfacea Chlorella virus 1, or ATCV-1, known to infect green algae. Green algae include more than 7,000 water-dwelling organisms that resemble plants but belong to a separate biologic kingdom. They are commonly found in aquatic environments like ponds, lakes and the ocean.
