Introduction Since the first immunoassays were developed in the 1950's the variety of assay formats has increased dramatically.

This new method, termed immuno-polymerase chain reaction (immuno-PCR), benefits from the specificity of antibodies and the sensitivity of PCR. Several independent studies undertaken since the publication of this seminal paper have consistently demonstrated the advantages of this technique over traditional ELISA, some achieving more than a 50,000 fold increase in sensitivity over the equivalent immunoassay [2]. Furthermore, immuno-PCR can be used in a multiplex format and offers accurate quantitation.

Microbiological diagnoses based on laboratory testing directly affect patient care and outcomes, including hospital infection control, duration of hospitalization, and laboratory efficiency, according to new guidelines from the Infectious Diseases Society of America (IDSA) and the American Society for Microbiology.

The new recommendations, which address appropriate and accurate use of laboratory tests to diagnose infectious diseases (IDs), were published online July 10 in Clinical Infectious Diseases by Ellen Jo Baron, PhD, from the Department of Pathology, Stanford University School of Medicine, and Cepheid, R&D, Sunnyvale, California, and colleagues.

We have developed a novel approach to categorize immunity in patients that uses a combination of whole blood flow cytometry and hierarchical clustering.

Conclusions

Comprehensive multi-factorial immune analysis resulting in immune profiles were prognostic, uncovered relationships among immune markers and identified a potential biomarker for the prognosis of cancer. Immune profiles may be useful to streamline evaluation of immune modulating therapies and continue to identify immune based biomarkers.

Both the treatment of patients with allergic diseases and the study of allergic disease mechanisms depend on a wide variety of assays that in various ways assess the presence and function of IgE antibody. The study of allergic diseases could benefit from the study of its 2 principle cellular participants, mast cells and basophils, but the basophil is more accessible than mast cells for ex vivo studies. Its functionality is tested by using 2 predominant methodologies: the secretion of mediators of allergic inflammation and the expression of proteins on the plasma membrane after stimulation. Each approach has benefits. There are also many operational details to consider regardless of which general approach is taken, and proper interpretation of the methods requires a good understanding of the reagents used and the receptors expressed on basophils and a detailed understanding of the factors regulating aggregation of cell-surface IgE.

of course, we lost cytologist's competencies... to look at cells.

We have to rely on secretions assays or cytometric analysis which work pretty well in our experience. Unfortunately, clinical data to correlate biological are not easy to collect and the topic is not very fashionable, somehow outdated.

But, if JACI is devoting a review on the topic, the tests might come back on stage?

Aydogan Ozcan, a professor of electrical engineering and bioengineering at the UCLA Henry Samueli School of Engineering and Applied Science, and his team have created a portable smartphone attachment that can be used to perform sophisticated field testing to detect viruses and bacteria without the need for bulky and expensive microscopes and lab equipment. The device weighs less than half a pound.

The XV International Congress of Immunology (ICI) held in Milan, Italy, August 22–27, 2013 is likely a perfect occasion to celebrate a marriage that was officiated in the mid-1970s, when monoclonal antibodies (mAbs) were created and became available for research. After a very brief period of engagement, the “official exchange of rings” was represented by the possibility to conjugate mAbs with adequate fluorochromes. This changed almost completely the main use of an instrument, that is, the flow cytometer, that had been created and commercialized few years before for the measurement of cellular DNA by fluorescent intercalating dyes such as ethidium bromide. The recognition of immune cells stained with fluorescent mAbs, typically performed by fluorescence microscopy, was then passed to complex (and at that time quite esoteric) instruments that could analyze thousands of cells in a few seconds, and could provide informative and highly repetitive results.
