Researchers uncover new piece of the HIV puzzle | Mucosal Immunity | Scoop.it

New research has revealed that a key immune system component -- innate lymphoid cells (ILC) -- is destroyed during acute HIV infection. This may play a key role in understanding the progression of the disease from HIV to AIDS.

Mucosal Immunology - Abstract of article: A dendritic cell targeted vaccine induces long-term HIV-specific immunity within the gastrointestinal tract

Mucosal Immunology is the official publication of the Society of Mucosal Immunology (SMI). It aims to provide a forum for both basic and clinical scientists to discuss all aspects of immunity and inflammation involving mucosal tissues. The journal reflects the interests of scientists studying gastrointestinal, pulmonary, nasopharyngeal, oral, ocular, and genitourinary immunology through the publication of original research articles, scholarly reviews, and timely commentaries, editorials and letters.
Gilbert C FAURE's insight:

Despite significant therapeutic advances for HIV-1 infected individuals, a preventative HIV-1 vaccine remains elusive. Studies focusing on early transmission events, including the observation that there is a profound loss of gastrointestinal (GI) CD4+ T cells during acute HIV-1 infection, highlight the importance of inducing HIV-specific immunity within the gut. Here we report on the generation of cellular and humoral immune responses in the intestines by a mucosally administered, dendritic cell (DC) targeted vaccine. Our results show that nasally delivered α-CD205-p24 vaccine in combination with polyICLC, induced polyfunctional immune responses within naso-pulmonary lymphoid sites that disseminated widely to systemic and mucosal (GI tract and the vaginal epithelium) sites. Qualitatively, while α-CD205-p24 prime-boost immunization generated CD4+ T-cell responses, heterologous prime-boost immunization with α-CD205-p24 and NYVAC gag-p24 generated high levels of HIV-specific CD4+and CD8+ T cells within the GI tract. Finally, DC-targeting enhanced the amplitude and longevity of vaccine-induced immune responses in the GI tract. This is the first report of a nasally delivered, DC-targeted vaccine to generate HIV-specific immune responses in the GI tract and will potentially inform the design of preventative approaches against HIV-1 and other mucosal infections.

Increased frequency of circulating Tc22/Th22 cells and polyfunctional CD38− T cells in HIV-exposed uninfected subjects : Scientific Reports | Mucosal Immunity | Scoop.it
Some individuals are resistant to HIV-1 infection despite repeated exposure to the virus, suggesting the presence of a complex antiviral response. Innate factors like IL-22 exert gut mucosal protection and polyfunctional T cells have been associated with low progression in HIV infection; therefore, we evaluated the frequencies of CD4+ and CD8+ T cell-secreting cytokines, including Tc22/Th22 cells and polyfunctional T cells in HIV-1-exposed uninfected individuals (EUs), their HIV-1-infected partners and healthy controls.

Gilbert C FAURE's insight:

These results suggest a protective role for mucosal HIV-1 envelope-specific IgA responses in the context of postnatal virus transmission. This finding supports further investigations into the mechanisms by which mucosal IgA reduces risk of HIV-1 transmission via breast milk and into immune interventions aimed at enhancing this response.

Gilbert C FAURE's insight:

Fichorova and colleagues analyzed the impact of infection and hormonal contraceptives (HCs) within a cohort of 832 women enrolled in Uganda and Zimbabwe. Participants were divided into groups based on the use of injectable DMPA, combined oral contraceptives (COC) or non-HC alternatives, with women recently infected with HIV (n = 199) compared with those who remained uninfected (n = 633) during a 6-month follow-up. Cervicovaginal infections (CVI) were diagnosed using PCR, antibody enzyme-linked immunosorbent assay (ELISA), wet mount and Nugent scoring. Samples collected via cervical Dacron swabs also were processed and simultaneously measured for the presence of eight cervical immunity biomarkers.

Higher Frequency of NK and CD4 + T-Cells in Mucosa and Potent Cytotoxic Response in HIV Controllers

Higher Frequency of NK and CD4 + T-Cells in Mucosa and Potent Cytotoxic Response in HIV Controllers | Mucosal Immunity | Scoop.it
HIV infection induces immune alterations, mainly in gut mucosa, where the main target cells reside. However, the evolution of the infection is variable among infected individuals, as evidenced by HIV controllers who exhibit low or undetectable viral load in the absence of treatment. The aim of this study was to evaluate the frequency, phenotype and activity of T and NK cells in peripheral blood and gut mucosa in a cohort of Colombian HIV controllers. Blood and gut biopsies were included. The fr
