COLUMBUS, Ohio – Research led by investigators at The Ohio State University Wexner Medical Center provides new hope for recovery from degenerative neurological diseases — such as ALS and multiple sclerosis — as well from as damage caused by traumatic brain and spine injuries and stroke.

Using a mouse model, researchers at Ohio State and the University of Michigan discovered a new type of immune cell that not only rescues damaged nerve cells from death, but partially reverses nerve fiber damage. The research team also identified a human immune cell line, with similar characteristics, that promotes nervous system repair.

Study findings are published in the journal Nature Immunology.

Since the advent of the European Union (EU) General Data Protection Regulation (GDPR) in 2018, the biomedical research community has struggled to share data with colleagues and consortia outside the EU, as the GDPR limits international transfers of personal data. A July 2020 ruling of the Court of Justice of the European Union (CJEU) reinforced obstacles to sharing, and even data transfer to enable essential research into coronavirus disease 2019 (COVID-19) has been restricted in a recent Guidance of the European Data Protection Board (EDPB). In this article, the authors examine whether there is room under the GDPR for EU biomedical researchers to share data from the EU with the rest of the world to facilitate biomedical research. They then propose solutions for consideration by either the EU legislature, the EU Commission, or the EDPB in its planned Guidance on the processing of health data for scientific research. Finally, they urge the EDPB to revisit its recent Guidance on COVID-19 research.

University of Groningen scientists created a new enzyme that can catalyse (speed up) an important reaction in organic chemistry. They added a copper complex to a protein that had no enzymatic properties. Next, they inserted an unnatural amino acid into the protein. Together with the copper, a side chain of the amino acid was able to catalyse the required reaction. The method to add unnatural groups to a protein could be used to design many other new enzymes. These could replace standard chemical catalysis and thereby contribute to making chemistry cleaner and more energy efficient.

The new generation CRISPR technology allows the restoration of the retinal and visual function after gene therapy in the treatment of a wide range of inherited ocular diseases. Using a mouse model with Leber Congenital Amaurosis (LCA) due to a clinically relevant pathogenic mutation in the Rpe65 gene, the team from the University of California at Irvine has successfully demonstrated the therapeutic potential of base editing for the treatment of LCA and by extension other blinding hereditary diseases. The researchers overcame some of the obstacles to the CRISPR-Cas9 system, such as unpredictable off-target mutations and low editing efficiency, by using cytosine and adenine base editors. The use of these editors allowed them to correct point mutations in a precise and predictable manner while minimizing unintentional mutations that could cause undesirable side effects. The basic editing treatment restored retinal and visual function in LCA mice to near normal levels. These results are extremely encouraging and represent a major step forward in the development of treatments for inherited retinal diseases.

The ‘D614G’ mutation (Aspartate-to-Glycine change at position 614) of the SARS-CoV-2 spike protein has been speculated to adversely affect the efficacy of most vaccines and countermeasures that target this glycoprotein, necessitating frequent vaccine matching. Virus neutralisation assays were performed using sera from ferrets which received two doses of the INO-4800 COVID-19 vaccine, and Australian virus isolates (VIC01, SA01 and VIC31) which either possess or lack this mutation but are otherwise comparable. Through this approach, supported by biomolecular modelling of this mutation and the commonly-associated P314L mutation in the RNA-dependent RNA polymerase, the authors have shown that there is no experimental evidence to support this speculation. They additionally demonstrate that the putative elastase cleavage site introduced by the D614G mutation is unlikely to be accessible to proteases.

After receiving an especially aggressive combination of antiretroviral (ARV) drugs and nicotinamide (vitamin B3), the man, who asks to be referred to as the São Paulo Patient to protect his privacy, went off all HIV treatment in March 2019 and has not had the virus return to his blood.

An international collaboration of researchers from the RIKEN Center for Biosystems Dynamics Research (BDR) in Japan and the Gladstone Institutes in the USA have generated 3D blastocyst-like structures from stem cells. Published in the journal Stem Cell Reports, the study shows that the blastocyst-like structures very closely resemble actual blastocysts, and even induce proper changes in the uterus after being implanted in pseudo-pregnant mice.
