Charged ultrafiltration membranes combined with optimal operating conditions can be used to enable the purification of monoclonal antibodies. Researchers at BERG-iBB (Alexandra Wagner, Marilia Mateus and Ana Azevedo) and BSIRG-IBB (Ana Ferraria and Ana Maria Botelho do Rego) have studied the influence of polyethersulfone membrane modification with polyethyleneimine and operating conditions on the removal of impurities from a mAb-containing cell culture fluid. High purity was consistently observed at pH 9 due to a simultaneous decrease in the transmission of soluble protein and increase in both IgG transmission and adsorptive capacity of the membranes. Declining wild fish stocks foster aquaculture industry for seafood supply. Such intensive fish rearing expedites disease transmission between individuals but vaccination can avoid feeding antibiotics to fish and economic losses. Plasmid DNA biopharmaceuticals research is an important BERG-iBB activity and fish vaccines development a challenge. Gabriel Monteiro, Marília Mateus and researchers from NIT-Rourkela, collaborated in designing, producing and characterizing vaccine candidates for immunizing fish against aeromoniases. DNA plasmids for expression of antigenic Aha1 peptides from A.hydrophila and a DNA nanoparticle-based delivery system were created/characterized. CHO cells transient transfection of the vaccine candidates was implemented, PLGA-chitosan nanoparticles ability for DNA complexation and protection from nucleases evaluated, and the system adequacy for up-scaled production discussed. Monoclonal antibodies are expensive to produce in part due to their purification process, which is centered in protein-A affinity chromatography. A recent publication by Ana Azevedo and co-workers from BERG-iBB highlights a novel purification strategy for mAbs comprising phenylboronic acid multimodal chromatography for capture followed by polishing by ion exchange monolithic chromatography and packed bed hydrophobic interaction chromatography. Process simulation and economical analysis show that the new process is worth investing in and could present a viable alternative to the platform process used by most industrial players. Luís Miguel raiado Pereira will be defending his Ph D thesis in Biotechnology at Instituto Superior Técnico, thrusday the 9th October 2014 (14:30 H, room PA3). During the last years, and under the supervision of Marília Mateus and Miguel Prazeres from BERG-IBB, Luís 

developed membrane hydrophobic interaction chromatography (HIC) membranes based on alkyl and phenyl ligands and studied their applicability  in the downstream processing of plasmid DNA.  The title of the thesis is “Hydrophobic Interaction Membrane Chromatography for the Downstream Processing of Plasmid DNA”.
