Detection of specific HPV subtypes responsible for the pathogenesis of genital warts

Detection of specific HPV subtypes responsible for the pathogenesis of genital warts | Virology News | Scoop.it

The aims of the study were to evaluate current strategies used for the monitoring of HPV infection in genital warts and to assess the suitability of laser-capture microdissection (LCM) as a technique to improve the understanding of the natural history of HPV types associated with genital wart lesions.

Methods

DNA and RNA were extracted from whole wart, surface swabs and LCM sections from 23 patients. HPV types present were determined using the Linear Array HPV Genotyping Test (Roche), with HPV DNA viral load and mRNA expression investigated using qPCR and qRT-PCR, respectively.

Results

Results indicated that swabbing the surface of warts does not accurately reflect potential causative HPV types present within a wart lesion, multiple HPV types being present on the surface of the wart that are absent in the lower layers of tissue isolated by LCM. Although it was shown that HPV DNA viral load does not directly correlate with HPV mRNA load, the presence of both DNA and mRNA from a single HPV type suggested a causative role in lesion development in 8/12 (66.6%) of patients analysed, with dual infections seen in 4/12 (33.3%) cases. HPV 6 and HPV 11 were present in more than 90% of the lesions examined.

Ed Rybicki's insight:

Interesting stuff: so, simply swabbing the surface of lesions does NOT give a good indication as to what is causing the lesion - and HPVs 6 and 11 are often present together in condylomas.

Study: Certain types of papilloma virus might prevent cervical cancer

Study: Certain types of papilloma virus might prevent cervical cancer | Virology News | Scoop.it
There are over 100 different types of human papilloma virus (HPV). Cervical cancer is known to be caused by infection with approximately 14 so-called "high-risk" types of this virus.

Researchers from Manchester looked at the different types of HPV found in cervical smears and invasive cervical cancers from HIV positive and HIV negative women in Kenya. They found high numbers of a specific type of HPV (type 53) in normal cervical smears from HIV positive women, but this was rarely found in HIV negative women. This sub-type was also never found in cervical cancers from either HIV positive or negative women.

Dr Ian Hampson, a Senior Lecturer in Viral Oncology from The University of Manchester who lead the study, said: "It is well known that HIV increases the number of different types of HPV found in any one patient which implies that HIV opens the door for infection with multiple types of HPV. If only high-risk types are present these will undoubtedly accelerate progression to cancer whereas if other types (eg type 53) are also present they may actually compete with the high-risk types to inhibit progression to cervical cancer."

Ed Rybicki's insight:

HPV PREVENTING cervical cancer??  An interesting thought - and undoubtedly eminently investigatable, given enough samples.  Hey, Anna-Lise??

Catholic board reconsidering HPV vaccine - Alberta Daily Herald Tribune

Catholic board reconsidering HPV vaccine - Alberta Daily Herald Tribune | Virology News | Scoop.it
Catholic board reconsidering HPV vaccine Alberta Daily Herald Tribune AHS Grande Prairie estimated that only 20-25% of students in Catholic schools receive the HPV vaccine at the Health Unit, compared with 95% of students in the public board, who...
Ed Rybicki's insight:

Don't Catholics get HPV?  Just like they can't get HIV? Really, the churches need to get their heads around the fact that people do not follow their doctrines very well.

Hi Ed...sadly I suspect that some Catholics do get HPV as well as HIV. But following your logic a rule or guideline should exist only if people follow it? That sounds like relativism at it worst. your logic we should do away with all laws that are not followed...hmmm. If you would like to understand better what the Church teaches I would be glad to try to help in that endeavor.
Ed Rybicki's comment, March 25, 2013 5:36 AM
I am a SERIOUSLY well-schooled ex-Catholic - so I will decline your offer. What I really would like to see is the Catholic Church take cognisance of the fact that their teachings around reproduction and the inhibition of it are completely out of touch with the modern world - and promote the use of infection control agents like condoms.
