Primary atopic disorders

Primary atopic disorders | Systemic Mastocytosis, Tinnitus etc | Scoop.it
Monogenic disorders have provided fundamental insights into human immunity and the pathogenesis of allergic diseases. The pathways identified as critical in the development of atopy range from focal defects in immune cells and epithelial barrier function to global changes in metabolism. A major goal of studying heritable single-gene disorders that lead to severe clinical allergic diseases is to identify fundamental pathways leading to hypersensitivity that can be targeted to provide novel therapeutic strategies for patients with allergic diseases, syndromic and nonsyndromic alike. Here, we review known single-gene disorders leading to severe allergic phenotypes in humans, discuss how the revealed pathways fit within our current understanding of the atopic diathesis, and propose how some pathways might be targeted for therapeutic benefit.

Diagnosis, classification and treatment of systemic mastocytosis - VJHemOnc

Diagnosis, classification and treatment of systemic mastocytosis - VJHemOnc | Systemic Mastocytosis, Tinnitus etc | Scoop.it
Systemic mastocytosis, though a rare disease, is more common than clinicians may think and diagnosis of the condition can often be missed. In this insightful interview, Deepti Radia, MBBS, MRCPI, FRCPath, of Guy’s and St Thomas’ Hospital NHS Foundation Trust, London UK, details the latest updates in the field, such as the new WHO classification guidelines for diagnosis and classification. Dr Radia also discusses the treatment options for this condition, which often consists of symptomatic treatment, but in cases of aggressive systemic mastocytosis there are now targeted treatments available, such as midostaurin, BLU-285 and DCC-2618. This video was recorded at the Myeloproliferative Neoplasms Advances Day 2017, London, UK.

What Is Aggressive Mastocytosis? | Systemic Mastocytosis, Tinnitus etc | Scoop.it
Mastocytosis is a rare disorder characterized by abnormal accumulations of mast cells in the skin, bone marrow, and internal organs (liver, spleen, gastrointestinal tract and lymph nodes). Cases beginning during adulthood tend to be chronic and involve the bone marrow in addition to the skin, whereas, during childhood, the condition is often marked by skin manifestations with no internal organ involvement and can often resolve during puberty.

FGFR1 translocation with concurrent myeloproliferative neoplasm, systemic mastocytosis, and lymphoblastic lymphoma: a case report | Systemic Mastocytosis, Tinnitus etc | Scoop.it
FGFR1 translocation may cause myeloid or lymphoid neoplasm but rarely systemic mastocytosis
(SM). Conversely, SM is associated with myeloproliferative neoplasm (MPN), but rarely
lymphoblastic lymphoma (LBL) or FGFR1 translocation. We report the first case of FGFR1
translocation in a patient with concurrent LBL, MPN, and SM. A 21-year-old male patient
presented with diffuse lymphadenopathies and leukocytosis. TdT+/cytoCD3+/CD79aweakly+
LBL was identified in the lymph node. Bone marrow had MPN, SM, and TdT+/CD79a+/cytoCD3weakly+
LBL.
