Abstract: Immunotherapy has been a mainstay of treatment for metastatic renal cell carcinoma (mRCC) since the introduction of high-dose interleukin-2. Recently, improved knowledge of immune regulation and tumor-host immune interactions has led to the development of several novel immunotherapies. Immune checkpoint blockade showed promise in early clinical trials - eventually leading to FDA approval of nivolumab (anti-PD-1) as a second-line treatment for mRCC. Despite encouraging results, PD-1 blockade alone does not achieve durable responses in the majority of patients treated. Improved biomarkers for patient selection, tumor vaccines and combination therapy may augment the efficacy of existing immunotherapies. This review summarizes recent progress in immunotherapy for RCC, focusing on a discussion of emerging agents.

The inhibition of myostatin signaling increases bone remodeling, bone mass and muscle mass, and it may provide a target for the treatment of both sarcopenia and osteoporosis. As myostatin is involved in osteoclast formation and bone destruction in rheumatoid arthritis, myostatin may be a target myokine for the treatment of accelerated bone resorption and joint destruction in rheumatoid arthritis. Numerous other myokines, including transforming growth factor-β, follistatin, insulin-like growth factor-I, fibroblast growth factor-2, osteoglycin, FAM5C, irisin, interleukin (IL)-6, leukemia inhibitory factor, IL-7, IL-15, monocyte chemoattractant protein-1, ciliary neurotrophic factor, osteonectin and matrix metalloproteinase 2, also affect bone cells in various manners. However, the effects of myokines on bone metabolism are largely unknown. Further research is expected to clarify the interaction between muscle and bone, which may lead to greater diagnosis and the development of the treatment for muscle and bone disorders, such as osteoporosis and sarcopenia.

Here's a scary insight for you.  If you have been taking vitamin E and or selenium.I don't know about you, but generally I don't pay much attention to these sort of studies because next week.  Some will come out to say that vitamin E is good, beats cancer, selenium beats cancer all to hell.So you have to decide for yourself.  I have an opinion, but I'm not gonna tell you it.

If you are still at the stage where you are decideing what treatment may be best for you read this. sometimes I tink that doctors keep some of the side effects secret. I wonder why? One thing that is passed over quickly if mentioned at all is incontinance from radiation. It is a BIG side effect. I don't mean pissing yourself I mean shitting. 6 or 7 times a day. You go out and you are always on the lookout for the nearest washroom....
