Relative expressions of structural genes and a number of transcription factors of the anthocyanin pathway relevant in Vaccinium species, and related key enzyme activities were compared with the composition and content of metabolites in skins of ripe fruits of wild albino and blue bilberry (Vaccinium myrtillus) found in Slovenia. Compared to the common blue type, the albino variant had a 151-fold lower total anthocyanin and a 7-fold lower total phenolic content in their berry skin, which correlated with lower gene expression of flavonoid 3-O-glycosyltransferase (FGT; 33-fold), flavanone 3-hydroxylase (FHT; 18-fold), anthocyanidin synthase (ANS; 11-fold), chalcone synthase (CHS, 7.6-fold) and MYBPA1 transcription factor (22-fold). The expression of chalcone isomerase (CHI), dihydroflavonol 4-reductase (DFR), leucoanthocyanidin reductase (LAR), anthocyanidin reductase (ANR) and MYBC2 transcription factor was reduced only by a factor of 1.5–2 in the albino berry skins, while MYBR3 and flavonoid 3’,5’-hydroxylase (F3’5’H) were increased to a similar extent. Expression of the SQUAMOSA class transcription factor TDR4, in contrast, was independent of the color type and does therefore not seem to be correlated with anthocyanin formation in this variant. At the level of enzymes, significantly lower FHT and DFR activities, but not of phenylalanine ammonia-lyase (PAL) and CHS/CHI, were observed in the fruit skins of albino bilberries. A strong increase in relative hydroxycinnamic acid derivative concentrations indicates the presence of an additional bottleneck in the general phenylpropanoid pathway at a so far unknown step between PAL and CHS.

Crown rot ( Phytophthora cactorum ) causes significant economic losses in strawberry production. The best control strategy would be to use resistant cultivars, but polygenically inherited resistance makes the breeding of the garden strawberry ( Fragaria × ananassa ) challenging. The diploid wild strawberry Fragaria vesca Hawaii 4 genotype was shown previously to have resistance against crown rot. To explore the resistance mechanisms, we inoculated the roots of Hawaii 4 with P . cactorum in a novel in vitro hydroponic system to minimize interference caused by other microbes. Major reprogramming of the root transcriptome occurred, involving 30% of the genes. The surveillance system of the plant shifted from the development mode to the defense mode. Furthermore, the immune responses as well as many genes involved in the biosynthesis of the defense hormones jasmonic acid, ethylene and salicylic acid were up-regulated. Several major allergen-like genes encoding PR-10 proteins were highly expressed in the inoculated plants, suggesting that they also have a crucial role in the defense responses against P . cactorum . Additionally, flavonoids and terpenoids may be of vital importance, as several genes involved in their biosynthesis were up-regulated. The cell wall biosynthesis and developmental processes were down-regulated, possibly as a result of the down-regulation of the key genes involved in the biosynthesis of growth-promoting hormones brassinosteroids and auxin. Of particular interest was the expression of potential resistance genes in the recently identified P . cactorum resistance locus RPc-1 . These new findings help to target the breeding efforts aiming at more resistant strawberry cultivars.

200 aromiaineesta makunsa saava mesimarja sopisi hyvin Suomen arktisiin olosuhteisiin, mutta sen laajamittaisempaan kasvatukseen ei ole onnistuttu löytämään vielä ratkaisuja. Kauhavalta löytyy kuitenkin yksi sitkeä marjantuottaja, joka on kokeillut mesimarjan viljelyä jo yli 20 vuotta.

The objectives of this research were a) to detect potential sources of primary inoculum of downy mildew of cultivated blackberry and b) to determine some components of resistance to P. sparsa in 4 wild blackberry species from Michoacan Mexico. A region-wide sampling of symptomatic and asymptomatic blackberry plants from nurseries, commercial plots, wild blackberries (R. adenotrichos) and rose plants (Rosa sp.), was conducted. Nested-PCR assays with PS3/PS1 primers in the first reaction followed by a second amplification with PR3/PR4 primers, amplifying a final fragment of 477 bp of the ITS region were used, and then sequenced in both directions. Detached leaflets of 4 common wild blackberry species were inoculated with P. sparsa sporangia under controlled conditions. Sporangiophores (SP) were recorded over a period of 18 days and oospores (OO) were counted 21 days after inoculation; the latent period (LP) and disease severity (DS) were also estimated. Phylogenetic reconstruction with Bayesian inference confirmed the presence of P. sparsa in all collected samples. Rubus adenotrichos, R. pringlei and R. sapidus were high inoculum producers, whereas R. cymosus allowed neither SP nor OO production and had the longest LP and lowest DS. Together, these results indicate that the primary source of inoculum for commercial plantings is mainly the blackberry plants from nurseries, and potentially 3 species of wild blackberries. Rubus cymosus has potential to be used in blackberry breeding programs as a source of resistance to P. sparsa. Mesimarjan viljely puutarhoissa on edelleen kokeiluasteella, vaikka sen viljelymahdollisuuksia on tutkittu jo 1600-luvulta lähtien. Kauhavalaisella marjatilalla mesimarjan pylväsviljelyä kokeillaan kasvutunnelissa. Arktisen marjan herkullisuus tulee sen sisältämistä yli 200 eri aromiaineesta. Luonnossa mesimarja on käynyt harvinaiseksi.

Baltian maista ei pitäisi tuoda Suomeen koristekasveja ja hedelmäpuita, Elintarviketurvallisuusvirasto Evira muistuttaa. Syy on tulipolte-niminen kasvitauti, jota esiintyy muissa Euroopan maissa mutta ei vielä Suomessa. Tauti todettiin Virossa kaksi vuotta sitten, ja se voi levitä Suomeen puutarhaharrastajien ostamien taimien mukana.
