Investigators for the nationwide trial, NCI-MATCH: MolecularAnalysis for Therapy Choice (EAY131), announced today at the annual meeting of the American Society of Clinical Oncology (ASCO) in Chicago that the precision medicine trial will open to patient enrollment in July. The trial seeks to determine whether targeted therapies for people whose tumors have specific gene mutations will be effective regardless of their cancer type. NCI-MATCH will incorporate more than 20 different study drugs or drug combinations, each targeting a specific gene mutation, in order to match each patient in the trial with a therapy that targets a molecular abnormality in their tumor. The study was co-developed by the National Cancer Institute (NCI), part of the National Institutes of Health, and the ECOG-ACRIN Cancer Research Group, part of the NCI-sponsored National Clinical Trials Network (NCTN). It is being led by ECOG-ACRIN.


NCI-MATCH is a phase II trial with numerous small substudies (arms) for each treatment being investigated. It will open with approximately 10 substudies, moving to 20 or more within months. The study parameters for the first 10 arms are being sent to 2,400 participating sites in the NCTN for review in preparation for patient enrollment beginning in July. The exact date for the opening of patient enrollment will be decided shortly after the ASCO meeting. Additional substudies are in development and will be added over time as the trial progresses.

A study using DNA testing offers perhaps the most credible evidence to date of adulteration, contamination and mislabeling in the herbal supplement industry.

 

Herbal products available to consumers in the marketplace may be contaminated or substituted with alternative plant species and fillers that are not listed on the labels. According to the World Health Organization, the adulteration of herbal products is a threat to consumer safety. Our research aimed to investigate herbal product integrity and authenticity with the goal of protecting consumers from health risks associated with product substitution and contamination.

 

A team of researchers used DNA barcoding to conduct a blind test of the authenticity for (i) 44 herbal products representing 12 companies and 30 different species of herbs, and (ii) 50 leaf samples collected from 42 herbal species. Their laboratory also assembled the first standard reference material (SRM) herbal barcode library from 100 herbal species of known provenance that were used to identify the unknown herbal products and leaf samples. They recovered DNA barcodes from most herbal products (91%) and all leaf samples (100%), with 95% species resolution using a tiered approach (rbcL + ITS2). Most (59%) of the products tested contained DNA barcodes from plant species not listed on the labels. Although they were able to authenticate almost half (48%) of the products, one-third of these also contained contaminants and or fillers not listed on the label. Product substitution occurred in 30/44 of the products tested and only 2/12 companies had products without any substitution, contamination or fillers. Some of the contaminants they found pose serious health risks to consumers.

 

Conclusion: Most of the herbal products tested were of poor quality, including considerable product substitution, contamination and use of fillers. These activities dilute the effectiveness of otherwise useful remedies, lowering the perceived value of all related products because of a lack of consumer confidence in them. The research team suggests that the herbal industry should embrace DNA barcoding for authenticating herbal products through testing of raw materials used in manufacturing products. The use of an SRM DNA herbal barcode library for testing bulk materials could provide a method for 'best practices' in the manufacturing of herbal products. This would provide consumers with safe, high quality herbal products.

Researchers at Weill Cornell Medical College have developed a vaccine that could help existing smokers quit for good and prevent those yet to try cigarettes from ever becoming addicted. The vaccine turns the recipient’s kidney into a factory continuously churning out antibodies that clear the bloodstream of nicotine before it has a chance to reach the brain and deliver it’s addictive rush. Unlike previously tested nicotine vaccines that only last a few weeks, the effects of a single dose of this new vaccine should last a lifetime.

 

Generally, there are two types of vaccines. The first are active ones that activate a lifetime immune response by presenting a bit of the foreign substance, such as a piece of virus, to the immune system. Nicotine isn't suitable for active vaccines as the molecule is too small to be recognized by the immune system. The second type is a passive vaccine, which delivers ready-made antibodies to an individual. While passive nicotine vaccines have been tested, they have failed in clinical trials because they only last a few weeks and require repeated, expensive injections.

 

Building on previous work on a new, third kind of vaccine initially tested in mice to treat certain eye diseases and tumor types, known as a genetic vaccine, the research team at Weill Cornell took the genetic sequence of an engineered nicotine antibody and put it into an adeno-associated virus (AAV), which is a virus engineered not to be harmful. They also included information that directed the vaccine to go to hepatocytes, which are liver cells. The antibody's genetic sequence then inserts itself into the nucleus of hepatocytes, and these cells start to churn out a steady stream of the antibodies, which neutralize the nicotine as soon as it enters the bloodstream.

The number of new cases of cancer in the world is rising, according to a new report that looked at cancer in 118 countries. Globally, the number of new cancer cases increased from 8.5 million in 1990 to 14.9 million in 2013, the study found. The world population rose from 5.3 billion to 7.1 billion during that time. In addition, cancer is accounting for an increasingly greater proportion of deaths: In 1990, 12 percent of all deaths in the countries studied were due to cancer, but in 2013, it was 15 percent.

 

The researchers specifically looked at 28 different types of cancer, and found that cases from nearly all of these types of cancer have increased in the last two decades — ranging from a 9 percent increase in cervical cancer cases to a 217 percent increase in prostate cancer cases. The only cancer that decreased during the study period was Hodgkin's lymphoma, which saw a 10 percent decrease in the number of new cases between 1990 and 2013.

 

The overall rise in cancer cases is partly due to longer life spans, since the risk of cancer increases with age. "With life expectancy increasing globally, the future burden of cancer will likely increase," the researchers said. The growing global population, increases in obesity and poor dietary habits also have contributed to the rise, they said.

 

Cancer is more common in men than in women, with 1 in 3 men worldwide developing cancer before age 79, compared with 1 in 5 women. The most common cancer overall was cancer of the lungs, trachea or bronchus, with 1.8 million new cases and 1.6 million deaths in 2013, followed by breast cancer and colon cancer. The most common cancer in men was prostate cancer, and the most common cancer in women was breast cancer.

 

A particularly concerning trend is an increase in cancer cases in developing countries, the researchers said. In 2013, the rates of new cancer cases were higher in developing countries than in developed countries for stomach cancer, liver cancer, esophageal cancer, cervical cancer, mouth cancer, and nose and throat cancer.

Scientists have developed a small-molecule-inhibiting drug that in early laboratory cell tests stopped breast cancer cells from spreading and also promoted the growth of early nerve cells called neurites.

 

The inhibitor overcomes a number of previous scientific challenges by precisely targeting a single component of a cell signaling protein complex called Rho GTPases. This complex regulates cell movement and growth throughout the body. Miscues in Rho GTPase processes are also widely implicated in human diseases, including various cancers and neurologic disorders.

 

Because the role of Rho GTPases in cellular processes and cancer formation is well established, researchers have spent years trying to identify safe and effective therapeutic targets for specific parts of the protein complex. In particular, scientists have focused on the center protein in the complex called RhoA, which is essential for the signaling function of the complex. In breast cancer for example, increased RhoA activity makes the cancer cells more invasive and causes them to spread, while a deficiency of RhoA suppresses cancer growth and progression.

 

Despite this knowledge, past efforts to develop an effective small-molecule inhibitor for RhoA have failed, explained Zheng, who has studied Rho GTPases for over two decades. Most roadblocks stem from a lack of specificity in how researchers have been able to target RhoA, a resulting lack of efficiency in affecting molecular processes, problems with toxicity, and the inability to find a workable drug design.
