Novel research has investigated the effect of dimethyloxalylglycine (DMOG) on cell metabolism. MitoXpress® Xtra probes, as available from AMSBIO, allowed the real-time analysis of cellular respiration and mitochondrial function. This analysis was performed on a 3D spheroid culture, produced from reagents also provided by AMSBIO to show impaired deep cell deoxygenation in tumours, ischaemic, and inflamed tissues. A new research in the field of skin tissue engineering has been published. The biocompatibility and activity of various custom star-PEG heparin hydrogels was tested. A. Watarai and her group were able to optimise the adhesion and proliferation of these cells. In addition to this, slight adjustment of hydrogen-delivered growth factors resulted in differentiation of fibroblasts into myofibroblasts. Recently a study investigating the pathology of lymphangioleiomyomatosis (LAM) and the relation to wild type mesenchymal cells was published. Lymphangioleiomyomatosis is a rare disease leading to unknown accumulation in the lungs and cysts formation. Among other analysing techniques fibroblast interaction with LAM cells was examined using basement membrane extract (BME provided by AMSBIO) as a 3D cell culture mimicking the in-vivo conditions. 3D Triculture Model for Evaluating Breast Cancer Progression (developed from technology used in our spheroid assays) is part of a project to demonstrate feasibility for an advanced drug screening platform. The model shows that incorporating vascular and stromal cells with breast cancer tumor spheroids allows them to more closely mimic the extracellular environment, cellular architecture and behaviour of actual tumors; this triculture promotes growth, invasion and endothelial recruitment. My last post looked at gut organoids - an exciting new way to study the human intestine in culture. Gut organoids were developed by Hans Clevers and his team at the Hubrecht Institute for Stem Cell Research. Now a Professor of Molecular Genetics at Utrecht University, Clevers is a pioneer of 3D cell culture who is continuing to explore the wonderful world of stem cells within the gut.
