Effective peptide and protein delivery to the skin has received much attention in the pharmaceutical industry, with many companies developing a variety of delivery devices to force peptides and proteins into and across the epithelium of the skin. Despite these many attempts, effective delivery of high-molecular-mass compounds has at best been poor. The water-in-oil microemulsion system may overcome the water-impermeable barrier of the epidermis and allows for effective delivery of highly water-soluble molecules such as peptides and proteins following topical application.

Using Laser Technology, Aleksandr Ovsianikov from the Vienna University of Technology wants to create microstructures with embedded living cells. This project will be funded by an ERC Starting Grant.

The behavior of cells strongly depends on their environment. If they are to be researched and manipulated, it is crucial to embed them in suitable surroundings. Aleksandr Ovsianikov is developing a laser system, which allows living cells to be incorporated into intricate taylor-made structures, similar to biological tissue, in which cells are surrounded by the extracellular matrix. This technology is particularly important for artificially growing biotissue, for finding new drugs or for stem cell research.

Conventional excipients such as surfactants, cosolvents are often added to improve drug solubility for both oral and parenteral routes. Complexation with cyclodextrins or formulating oil in water emulsions are also employed to improve poorly soluble drugs. Today the growing demand to fabricate drug delivery systems capable of encapsulating high levels of active ingredients with additional benefits such as drug targeting capability has resulted in a plethora of technologies exploiting the unique physical properties of nano-sized structures. Nanotechnologies such as nano-crystals, solid lipid nanoparticles (SLN), nano-emulsion, polymeric nanoparticles, liposomes and polymeric self-assemblies have been employed to enhance drug solubilization. In this review, we will focus on four types; namely nano-crystals, SLN, liposomes and polymeric self-assemblies.
