Genetically engineered protein responds remotely to red light.

 

A team of biological engineers has developed a light-sensitive protein that permits scientists to control activity inside the brains of mice from outside the rodents’ skulls. The protein, called Jaws, promises to expand scientists’ ability to study brain activity in experimental animals and -- eventually -- humans. Ultimately, it holds the prospect of facilitating treatment of human conditions such as epilepsy.

 

Researchers are also using the protein to treat eye disease in experimental animals. Here, an immediate goal is therapy for certain eye ailments in humans.

 

Scientists use optogenetics, as the technology is known, to study the behavior and pathology of experimental animals’ brains by shining light on proteins known as opsins. Introduced into the brain aboard viruses, the opsins respond to the light by suppressing or stimulating electrical signals in brain cells. Optogenetic inhibition of the electrical activity of neurons enables the causal assessment of their contributions to brain functions. Red light penetrates deeper into tissue than other visible wavelengths. The red-shifted cruxhalorhodopsin, Jaws, derived from Haloarcula (Halobacterium) salinarum (strain Shark) and engineered to result in red light–induced photocurrents three times those of earlier silencers. Jaws exhibits robust inhibition of sensory-evoked neural activity in the cortex and results in strong light responses when used in retinas of retinitis pigmentosa model mice.

 

The opsins normally used in brain studies are sensitive to blue, green, or yellow light. Because bodily tissue absorbs those colors easily, the sources of such light must lie inside the brain. Typically, the light is delivered through an optical fiber implanted in an experimental animal’s brain. Jaws can noninvasively mediate transcranial optical inhibition of neurons deep in the brains of awake mice. The noninvasive optogenetic inhibition opened up by Jaws enables a variety of important neuroscience experiments and offers a powerful general-use chloride pump for basic and applied neuroscience.

 

A team led by Ed Boyden, associate professor of biological engineering and brain and cognitive sciences at the Massachusetts Institute of Technology, in Cambridge, reporting in Nature Neuroscience, demonstrated that red light shone from outside a mouse’s head can influence the Jaws protein up to three millimeters deep inside the brain. In fact, Boyden said, "we think the light goes further into the brain." A mouse’s brain is only about four millimeters thick.

 

"This is a huge advance, in that it allows for much deeper penetration of effective light," said David Lyon, an associate professor of anatomy and neurobiology at the University of California, Irvine School of Medicine. Lyon was not involved in the research on Jaws.

(Reuters) - Apple Inc is building a team of senior medical technology executives, raising hackles in the biotechnology community and offering a hint of what the iPhone maker may be planning for its widely expected iWatch and other wearable technology.

Over the past year, Apple has snapped up at least half a dozen prominent experts in biomedicine, according to LinkedIn profile changes. One prominent researcher moved two weeks ago, and Apple is recruiting other medical professionals and hardware experts, although the number of hires is not clear, said two people familiar with the hiring, who declined to be named.

Much of the hiring is in sensor technology, an area Chief Executive Tim Cook singled out last year as primed "to explode." Industry insiders say the moves telegraph a vision of monitoring everything from blood-sugar levels to nutrition, beyond the fitness-oriented devices now on the market.

Researchers at the J Craig Venter Institute recently unveiled their first self-replicating synthetic bacteria (M. mycoides JCVI-syn1.0) whose DNA was ‘programmed’ base pair by base pair. To verify that they had synthesized a new organism and not assembled the DNA from another natural bacteria, scientists encoded a series of ‘watermarks’ into the genes of M. mycoides JCVI-syn1.0. There are four of these hidden messages: an explanation of the coding system used, a URL address for those who crack the code to go visit, a list of 46 authors and contributors, and a series of famous quotes. The presence of these watermarks verifies that M. mycoides JCVI-syn1.0 truly is synthetic and demonstrates the precision and power of JCVI’s new techniques in synthetic biology.

Our entire lives, when you think about it, are built around rewards—the pursuit of money, fun, love, and tacos.

How we seek and respond to those rewards is part of what determines our overall happiness. Aristotle famously said there were two basic types of joy: hedonia, or that keg-standing, Netflix binge-watching, Nutella-from-the-jar selfish kind of pleasure, and eudaimonia, or the pleasure that comes from helping others, doing meaningful work, and otherwise leading a life well-lived.

Recent psychological research has suggested that this second category is more likely to produce a lasting increase in happiness. Hedonic rewards may generate a short-term burst of glee, but it dissipates more quickly than the surge created by the more selfless eudaimonic rewards.

"Happiness without meaning characterizes a relatively shallow, self-absorbed or even selfish life, in which things go well, needs and desire are easily satisfied, and difficult or taxing entanglements are avoided," a study in the Journal of Positive Psychology found last year.

In publishing its report on open access journals in the humanities and social sciences yesterday, the British Academy reminds us that the open access debate has a continuing tendency to concern itself with questions of the applicability of open access innovations to particular disciplines. Openness will require some definition before we proceed. One definition of openness might be ‘for the many, not the few’, as typified by notions of the public good. Open access has a strong case on these terms alone, irrespective of how limited any public utility might seem to those producing esoteric literature. Another definition might include ideas of multiple party involvement, in contrast to single party ownership. From this perspective, open access creates the potential for new utopias of collaboration and co-creation of knowledge. Thirdly, openness might simply be defined as the opposite of a certain form of ‘closed-ness’, in which form, content, innovation, communication, benefit and critique are closely circumscribed and controlled by a guild of experts. Such closed-ness is typical of lay criticism of the academy as irrelevant.

Each of these aspects has, at some point, encountered strong objections from particular voices within the academy when applied to research. While one ought not to be tempted to knock down straw men when invited to do so, rebuttals are often voiced by those wishing to compare and contrast the more discursive disciplines with those more deeply rooted in the scientific method. In treating each aspect of this definition in turn, we may start to see a bigger picture.

 

It is somewhat axiomatic that the intellectual pursuits of academics are mercifully free from questions of personal gain; indeed, it is argued that the purpose of the academy is to create the space to nurture such freedoms. However, it is also true that the driving force of self-interest is far from absent from the research world, ranging from the practices of attribution of knowledge discovery to individuals to the vagaries of Cornford’s 1908 Microcosmographia Academica. The process of knowledge dissemination straddles both worlds, in that it is through publication that one converts freely pursued research into personal attribution and credit. Though it is perhaps tempting to reduce the academy in this way, to do so entirely ignores the other side of the dissemination equation: the reader. To ignore the importance of the reader to the publication process is to reduce it to its most venal status, and academia itself becomes meanly extrapolated to a neat irrelevance.

 

Public funders of research, of course, cannot ignore the reader. We have the right to demand that the research we fund can fulfil its greatest potential relevance, and we have the right to place the burden of responsibility to achieve this on those being funded. Open access is an obvious way to do this, but in making research outputs more openly available, policymakers may entirely respect and preserve the symbiosis of academic freedom and credit: open access can (and I believe should) work within these parameters. HEFCE’s open access policy, with its generous exceptions, shows that it is possible to do this, and in this way, research may be more successfully conducted for the benefit of the many, as well as the few, and in ways that are entirely compatible with the economies of individual benefit.

Nearly 10 years ago, Vanderbilt University cognitive neuroscientist Randolph Blake and his postdoc Duje Tadin needed to give their study participants the experience of complete darkness. They were testing their new transcranial magnetic stimulator (TMS) and developing protocols for a series of experiments involving the generation of phosphenes—light experienced by subjects when there is none. So the researchers ordered high-end blindfolds, designed to block all light from reaching the eyes.

When the blindfolds arrived, Blake tried one out. “I can’t remember what prompted me to do it, but on a lark, I put them on myself first and waved my hand in front of my eyes,” he recalls, “and had this faint sense that I could see my hand moving.”

 

Tadin then tried it and had the same experience. The two replicated the mini experiment in the TMS lab, a small, dark room on the sixth floor. And again, both researchers could just barely see their hands through the blindfolds. “You could see this faint shadow, this faint impression of something moving back and forth in rhythm with your motions,” Blake says. But, when Blake waved his hand in front of Tadin’s blindfolded face, Tadin saw nothing. “That got us excited,” Blake says.

The duo traipsed around the building eagerly blindfolding their colleagues and asking them to report what they saw. “About half reported seeing something,” Blake says.

 

To test what was happening, however, the researchers knew they needed to come up with a better way to characterize what people were actually seeing. “What we discovered was an inherently subjective experience,” says Tadin. “There’s no easy way to ascertain that I’m telling the truth.” Unable to think of a reasonable way to measure the phenomenon, they set the project aside.

 

A few years later, running his own lab at the University of Rochester, Tadin told the story to graduate student Kevin Dieter, who encouraged Tadin to give the project another shot. They devised a conservative experimental setup in which they attempted to control the subjects’ expectations: they told study participants that one blindfold had little, imperceptible holes that might allow them to see through, while another blindfold would successfully keep out all light. (Both blindfolds were, in fact, totally lightproof.) A subject’s experience with the first blindfold could then guide his expectations for a second trial using the other blindfold. Specifically, if he had seen something with the first blindfold, he would certainly not expect to see anything with the second one. But even under these conditions, nearly 50 percent of subjects reported having at

 

least a “visual sensation of motion” while wearing the second blindfold.

The results hold “implications for how our different sensory systems work together,” Tadin says. Dealing only with subjective reports, however, still made him uneasy. So he turned to an eye-tracking device—used without the blindfolds but in complete darkness—to detect the movement of subjects’ eyes as they viewed the hand they reported seeing. People cannot move their eyes smoothly unless they have a visual target to lock on to, Tadin explains. If they just thought they saw their hand, jerky eye movements should reveal the truth.

Google gaat genetische en moleculaire informatie verzamelen van proefpersonen om te komen tot een volledig beeld van wat een gezond mens zou moeten zijn. De internetgigant hoopt op die manier medici te helpen bij het eerder opsporen van ziektes zoals hart-en vaatziekten en kanker.

 

De zoekgigant begint met het verzamelen van anonieme genetische en moleculaire informatie bij 175 proefpersonen, maar dit moeten er later duizenden worden, zo schrijft de Wall Street Journal. Van de proefpersonen worden onder andere lichaamsvochten zoals urine, bloed, speeksel en tranen verzameld.

Behandeling

Het Baseline project beperkt zich niet tot bepaalde ziekten, en zal honderden verschillende monsters verzamelen met behulp van een breed scala aan nieuwe diagnostische instrumenten. Google zal vervolgens haar enorme rekenkracht gebruiken om patronen en ‘biomarkers’ in de informatie te ontdekken. Deze biomarkers moeten medici helpen bij het veel eerder detecteren van een ziekte.

Uit het onderzoek kan bijvoorbeeld een ‘biomarker’ te voorschijn komen die sommige mensen helpt om vet voedsel efficiënt af te breken, waardoor ze minder risico lopen op een hoog cholestrol en hart- en vaatziekten. Mensen die deze eigenschap missen kunnen slachtoffer worden van hartaanvallen op jonge leeftijd. Wanneer Baseline een biomarker geïdentificeerd heeft, kunnen medici controleren of mensen deze missen en hen helpen hun gedrag aan te passen of een nieuwe behandeling ontwikkelen om ze beter te helpen.

De meeste ontdekte biomarkers zijn gerelateerd aan ziekten die zich in een laat stadium bevinden, omdat onderzoeken zich meestal focussen op zieke patiënten. Onderzoekers hebben met afwisselend resultaat geprobeerd deze te gebruiken om ziektes eerder op te sporen, zo vertelt Sam Gambhir, voorzitter van de Afdeling Radiologie aan de Stanford University Medical School die al ruim een jaar meewerkt aan Baseline.

Het project wordt geleid door Andrew Conrad, een 50-jarige moleculair bioloog, pionier in goedkope tests voor HIV in bloedplasma. Hij heeft een team van ongeveer 70 tot100 deskundigen tot zijn beschikking uit gebieden met inbegrip van fysiologie, biochemie, optica, beeldvorming en moleculaire biologie.

Conrad en Gambhir geven toe dat het project een grote sprong in het onbekende is, omdat het menselijk lichaam heel complex is en er maar weinig bekend is over het samenspel tussen DNA, enzymen en eiwitten en hoe omgevingsfactoren zoals voeding daar invloed op hebben. Het is mogelijk dat het project biomarkers oplevert die onderzoekers slechts weinig over ziekten vertellen. Conrad verwacht echter dat er in kleine stappen vooruitgang zal worden geboekt.

Hoewel Google laat weten dat de informatie van Baseline anoniem zal zijn en het gebruik beperkt blijft tot medische en gezondheidsdoeleinden, roept het toch privacy kwesties op. Zo kan de informatie van gigantische waarde zijn voor verzekeraars. Google benadrukt dat de informatie niet met verzekeraars wordt gedeeld. Het project wordt gecontroleerd door institutionele toetsingscommissies die toezicht houden op alle medische onderzoeken met mensen.

It is unclear what causes bipolar disorder - a condition characterized by dramatic changes in mood. But researchers from the University of Michigan Medical School have created the first stem cell model for bipolar disorder, which they say could uncover the origins of the condition and open the door to new treatments.

The research team recently published their study in the journal Translational Psychiatry.

 

To reach their findings, the investigators obtained skin samples from people with bipolar disorder, alongside skin samples from individuals without the condition.

By exposing small samples of skin cells to carefully controlled conditions, the researchers turned them into induced pluripotent stem cells (iPSCs). These are stem cells that have the potential to be turned into any other type of cell. The team then turned the iPSCs into neurons.

 

They measured gene expression of the iPSCs and then re-evaluated gene expression once the stem cells became neurons.

From this, the team found significant differences between the stem cells taken from bipolar patients and those taken from individuals without the condition.

Herken je dat? Het gevoel dat je geen aansluiting kan vinden bij anderen. Dat je er niet bij hoort hoe je ook je best doet. Hoe ga je op een 'normale' manier met mensen om? Het gevoel dat alles en iedereen tegen je is hebben we allemaal wel eens.

We kunnen pas spreken van een persoonlijkheidsstoornis als:

Iemands persoonlijke eigenschappen te extreem en hinderlijk worden. Iemands gedrag zo onberekenbaar of egocentrisch is dat de omgeving er last van heeft....en dat niet af en toe, maar vrijwel voortdurend. Er kan dan sprake zijn van een persoonlijkheidsstoornis. Je kunt hier een online test doen:

Een persoonlijkheidsstoornis is ook wel een stoornis in denken, voelen en handelen. Dan is het beeld dat je van jezelf of van anderen hebt verstoord. Je hebt een heel intens gevoel, of juist helemaal geen gevoel. Dit veroorzaakt moeilijkheden in de omgang met anderen. Impulsiviteit komt ook voor bij een persoonlijkheidsstoornis.

Een persoonlijkheidsstoornis heeft dus grote invloed op werk, gezin en sociale situaties en het is belangrijk voor je welzijn dat je hulp zoekt.
