Note: A striking example for the consequences of the introduction of MALDI-TOF for microbiology. Its just a few years ago that this bacterium was identified by molecular methods and later detected in humans by MALDi-TOF. There is now a slowly but constantly increasing number of human case reports and this now the first report for food.


FULL ABSTRACT:

Wohlfahrtiimonas chitiniclastica is an emerging zoonotic bacterium commensally living in larvae of particular flies. It has been associated with human and animal infections but never isolated from food. In the present study, a whole chicken carcass was rinsed in buffered peptone water which was then inoculated into BHI and the growth plated onto selective medium. Species identification was performed by MALDI-TOF MS. Those bacteria identified as W. chitiniclastica were subjected to 16S rRNA sequencing for confirmation and MEGA software was used to obtain their phylogenetic position. The findings of this study raise concerns regarding the abattoir, transport and stock practices of frozen meat carcasses and should be of interest with regard to microbiology, entomology and food production.

Conclusions: MALDI-TOF MS is useful for rapid identification of both yeast and mold cultures of P. marneffei and differentiation from related species. However, accurate identification to the species level requires database expansion using P. marneffei strains. Note: There is a nice passage in the paper worth to be cited: “With improvement of commercial databases, MALDI-TOF MS may replace genotypic methods as a cost-saving, first-line identification system for the less common fungal pathogens. Compared to other MALDI-TOF MS systems such as Vitek MS which also used for identification of various bacteria and fungi, the Bruker system has the advantage of being more versatile and allowing database enlargements [46]. Therefore, the Bruker system should be considered by laboratories which foresee the need of expanding databases for identifying less common pathogens such as P. marneffei.”
